Introduction
Notre équipe étudie comment les molécules de guidage axonal régulent les interactions cellulaires au cours du développement du système visuel et dans des maladies oculaires. Nous étudions aussi la fonction et l’évolution des projections commissurales.
Présentation
Depuis sa création, notre équipe étudie le développement des connexions neuronales dans le système nerveux central. Ce domaine de recherche est en pleine expansion car il a été montré que les mécanismes et les molécules qui régulent la croissance des axones et la migration des neurones, modulent aussi les interactions cellulaires en dehors du cerveau, dans de nombreux organes en développement et dans les tumeurs cancéreuses. Nos travaux visent principalement à comprendre la fonction et le mode d’action des molécules de guidage axonal au cours du développement du système nerveux, et en particulier du système visuel.Nous nous intéressons plus particulièrement à des molécules appelées Slit et Sémaphorines, ainsi qu’à leurs récepteurs respectifs, Robo et Plexines.
Nous travaillons sur un nombre limité de modèles (rétine, neurones précérébelleux, neurones commissuraux). Nous essayons ainsi de comprendre comment s’organisent les couches cellulaires dans la rétine, comment migrent et se différencient les cellules de crêtes neurales (qui forment le globe oculaire) et comment les axones commissuraux peuvent connecter les deux moitiés du cerveau. Pour répondre à ces questions nous utilisons divers modèles animaux (oiseaux, poissons, rongeurs, etc.) et notamment des lignées de souris transgéniques. Nous étudions les conséquences de l’inactivation des gènes Slit et Robo sur la neurogenèse, la formation de néo-vaisseaux et la tumorigenèse. Nous étudions également la régulation des interactions cellulaires au cours de la myélinisation et la rémyélinisation à l’aide de nouvelles méthodes génétiques et d’imagerie. Enfin nous essayons de développer des outils thérapeutiques permettant de régénérer la cornée et le nerf optique.
